Basically, it takes places at this hugely pristigious high school where a group of guys, known as the Host Club, are paid by girls (from the school) to hang out with (they usually just sit and drink tea at a table.) Of course, the guys are apparently incredibly hot, so the girls gladly pay.
Haruhi Fujioka, a high school girl accidentally walks into the Host Club's room one day, and Tamaki (the leader of the Club) immediately takes to Haruhi. Haruhi looks sort of mann'ish at this point, so Tamaki thinks she's a guy.
She gets a little freaked out by the Host Club, and out of nervousness, accidentally knocks over a billion dollar vase in the Host Club Room. To work off the money, she now must act as the Host Club's dog, buying refreshments for them and such.
Eventually the Club sees promise in Haruhi, and decides to give her a full makeover to look like a decent-looking guy, and maybe even a Host. Its at this point where they find out she's a girl, but after the makeover, Haruhi accepts the offer of becoming a Host, and they promise to keep her gender a secret.

That's the plotline in a nutshell. Also, throughout the anime, Tamaki and Haruhi's relationship slowly builds. :3

Its a great anime.
